Effect of BuShenYiQi tang on hyporesponsiveness of hypothalamo-pituitary-adrenal
axis (HPAA) in asthma model.

PURPOSE: To determine the effect of BuShenYiQi
tang on hypothalamo-pituitary-adrenal axis in asthma model.
METHODS: 40 BN mice were randomly divided into
4 groups. All animals were sensitized and challenged by ovalbumin for
7days and following 7days without challenge, then challenged for 7 days
again. Group BAC and D were administered by low, moderate and high
dose Chinese herbal formula BuShenYiQi tang respectively from day 21
to day 35, meanwhile, group A were administered with saline instead
of Chinese herbal formula. Plasma CORT was measured by enzyme-link immunosorbent
assay in all groups. Serum ACTH was detected by radioimmunoassay and
the _expression of CRHmRNA was measured by Real Time-PCR.
RESULTS: Asthma models show eosinophile granulocyte
inflammation. ACTH_ACRHmRNA in Chinese herbal groups is significantly
increased in a dose dependent matter than those of control group (p<0.05),
increase trend rather than significant difference of CORT in group D
was found.
CONCLUSIONS: BuShenYiQi tang can improve the
hyporesponsiveness of hypothalamo-pituitary-adrenal axis in dose dependent
matter, the target of it's effect lies in the hypothalamus or above
levels. We can concluded that BuShenYiQi tang can treat asthma by improving
the endogenious protective mechanism, it can reduce both the dose and
the side effects of inhaled glucocorticosteriods when administered to
asthma patients accompanied with inhaled glucocorticosteriods. it will
bring a high cost-effect value when it is used in clinical practice.
