Polysaccharides from Echinacea purpurea.

The investigation of immunomodulatory activity of
Echinacea purpurea led to the isolation of nine polysaccharides and
an arabinogalactan protein with immunostimulatory activity. Their molecular
weight of 1,600,000 -1,800,000 was determined on gel filtration PHLC
column monitored by RI detector with dextrans of know MW as calibrated
standard. The structures were identified by the neutral monosaccharide
analysis and methylation analysis. The polysaccharides consist of arabinose
and galactose at different ratios. With griess assay, we demonstrated
that these polysaccharides at the concentration of 1 Ð 8.5 _g/ml could
induce nitric oxide production from murine macrophage cell line, and
therefore they may function to modulate immune responses. The immunostimulatory
activity and the concentration variation of polysaccharides from the
fresh juice preparations of Echinacea purpurea were investigated
as well.
